Hi Colleen. Optimisation is where you use the "Save for Web" option, to reduce the quality of your images, to reduce the images' filesize, so they download faster on the web. Opacity is the opposite of Transparency. If you decrease the opacity of an image, it starts to become transparent. There's no such thing as an animated jpg as far as I know. Only animated gifs.
